聚焦人多能干细胞神经定向分化、体内命运追踪及功能神经环路重建。
基于多种大脑损伤动物模型,如脑卒中、机械损伤及阿尔茨海默氏症等中枢神经损伤与退行性疾病,系统开展人神经前体细胞移植策略的优化探索,并将细胞治疗与康复训练干预相结合,旨在通过细胞替代与微环境调控,修复受损神经组织,重建功能性神经环路,最终实现神经功能的持久恢复。在此基础上,我们拟利用先进的体内示踪技术及单细胞测序技术,对移植细胞在宿主脑内的存活、迁移、分化及整合情况进行长期动态监测,以期揭示其修复作用的细胞与分子机制,最终为中枢神经系统疾病的细胞治疗提供可靠的科学依据和临床策略,推动神经干细胞的临床应用转化。
