In this talk, Dr.Xiong will present a cryo-EM structure of the SARS-CoV S glycoprotein in complex with a neutralising antibody isolated from a SARS survivor, this structure reveals an un-expected spike protein conformation that represents a fusogenic intermediate state. This intermediate state is shown to be induced by antibody binding to the S receptor binding domain. Inspired by this structure, we confirmed, by a series of biochemical experiments, that binding of the receptor rather than pH change or proteolytic activation serves the fusion trigger for SARS-CoV and possibly other coronaviruses. This result also suggests antibodies target the S receptor binding region may be advantageous due to its potential of inducing premature fusion triggering
